Regeneration of high performance ion exchange chromatographic column in protein separations and purifications

英文摘要 | Irreversible adsorption of proteins on high-performance ion-exchange packings can result in column deterioration and damage. TSKgel CM-5PW column and a DEAE-SPW (75 x 7.5 mm I.D.) column fouled by irreversible adsorption of proteins were treated with 2% trypsin solution under 37 degrees C for 24 hours, the back pressure was significantly declined, the number of theoretical plate was increased by 4 to 13 times and ion-exchange capacity was restored. The extent of column regeneration was related with the volume and concentration of the trypsin solution. |
