英文摘要The mass transfer and chromatographic behavior of mouse serum immunoglobulin G(IgG) on protein G perfusion affinity chromatographic column were investigated experimentally. Experimental data have indicated that the protein G column had irreversible adsorption to IgG but no non-specific adsorption to the other proteins in serum under the investigation conditions. It was found that variations of the elution buffer composition and pH have played an essential effect to some extent on the retention time and the peak shape of IgG through changing the dissociation constant (K-D). The influence of mobile phase flow rate on the chromatographic behavior of IgG was also studied. The results indicated that the association/dissociation kinetics between IgG and protein G affinity packings became the rate-limiting step in perfusion affinity chromatographic separation process.
