Chemical speciation analysis of selenium in biological samples by a hyphenated technique of high performance liquid chromatography-inductively coupled plasma mass spectrometry

| 英文摘要 | A hyphenated technique for chemical speciation analysis of selenium in biological samples was described. A reversed ion-pair high performance liquid chromatograph is introduced for separating selenocompounds, at the meantime an inductively coupled plasma mass spectrometry was combined for on-line elemental determination. A series of standard compounds were used to achieve qualitative and quantitative analysis. Seven seleno-containing compounds, Na2SeO4, Na2SeO3, trimethyl selenonium iodide ( TmSeI ), selenocystaine ( SeCys ), selenourea ( SeUr ), selenomethionine ( SeMet ) and selenocystamine ( SeCM ), can be individually detected. The detection limits are 0.1-1.5 mu g/L, and the linear calibration range for Se concentrations is between 1 and 100 mu g/L. The method was also applied to analyze human liver cytosolic extracts, human serum, and human urine. Only 50 mu L of the samples are needed and the recoveries of spiked seleno-compounds are 93% -117%. The hyphenated method possesses the merits of simple operation, high sensitivity and good repeatability, which is a feasible method for speciation analysis of small molecular selenocompounds in different biological samples. |
