以六味地黄丸为例的中药制剂网络药理分析方法初步研究

中文摘要中药是我国传统文化的瑰宝,不但有独特的理论体系,而且在临床治疗方面有巨大贡献,因此现代科学对其研究也日益增多。但由于中药的成分复杂、靶点多样、作用途径不一等特点,使其在现代研究中遇到阻碍。随着网络药理学的诞生,药物研究从“单成分,单靶点” 模式转变为“多成分,多靶点” 模式。而网络药理学中的“药物—靶点—疾病”相互作用网络能够帮助研究人员从分子水平理解中药发挥作用的机制,也可以系统地揭示中药制剂中化学小分子对疾病的影响,因此中药制剂网络药理学可以作为中药现代化研究的切入点。 本课题以我国传统中药六味地黄丸为研究对象,利用六味地黄丸的化学成分数据库,蛋白质数据库和疾病数据库等多种资源,对六味地黄丸的化学成分进行了收集和整理,并对化学成分进行靶点预测,然后通过系统生物学方法构建六味地黄丸“化学成分—靶蛋白—疾病(基因)”的网络,以从分子水平阐释六味地黄丸治疗疾病的作用机制。并利用网络聚类和解构等分析技术对所建网络进行分析,结果显示六味地黄丸对治疗心血管疾病方面有较好的效果,并得到了文献验证。 综上所述,本文初步建立了一种高效快速的分析中药制剂网络药理的方法,为中药领域的现代化研究提供很好的研究思路。该方法具有较高的预测精度,可用于发现中药成分的潜在作用靶点,同时为研究中药的物质基础和分子作用机制提供了方法学参考。
英文摘要Traditional Chinese Medicine (TCM) is an excellent traditional cultural treasure. It not only has a unique theoretical system, but also makes tremendous contributions in the clinical treatment. Thus more and more studies about TCM are emerging in modern science. Due to the diversity of TCM compounds, the complexity of targets and the difference of metabolic pathways of medicine, the TCM research is hampered in modern society. Along with the emerging of network pharmacology, the medicine research mode turned from "single-component, single-target" to "multi-component, multi-target". The "drug - target - disease" interaction network can help us understand how TCM works in molecular level and how TCM impacts the disease process systematically. So TCM network pharmacology can be an entry point for modern TCM research. Liu-Wei-Di-Huang Wan (LWD) was used as the study case in this subject research. We used a variety of resources such as the compounds of herbal formulae, chemical molecular databases, protein databases and disease databases. We collected and organized LWD compounds data, predicted their targets and then constructed "LWD chemical compounds --targets--disease (gene)" network. So we can achieve better understanding of the mechanism in disease treatment for LWD at molecular level. Then we analyzed this network using clustering and deconstruction. The network demonstrated that LWD had a good effect for the treatment of cardiovascular diseases and the results were verified by literatures. In summary, this paper established a method for rapid and efficient analysis of network pharmacology of TCM preliminarily. It provided a good idea for modernization of TCM research. This method also had high prediction accuracy and can be utilized to identify potential targets of herbal formulae. The present approach may play an important role in studying the material basis and the molecular mechanism of TCM.
