英文摘要The thesis presents the principle, development, and applications of laser tweezers Raman spectroscopy (LTRS). Chapter 1 introduces the history and basic principle of Raman effect, the technique development and applications in biomedicine of Raman spectroscopy. Chapter 2 presents the principle of laser tweezers, configuration and calibration of laser tweezers Raman spectroscopy. Chapter 3 first introduces the experiment named as optical levitation and manipulation of stuck particles with pulsed optical tweezers, then presents our theoretical analysis and the numerical modeling to explain their experimental results. The result of our numerical modeling is qualitatively consistent with the experimental result. Chapter 4 presents the applications of LTRS in biomedicine. It includes two parts. The first part concerns study of the effect of alcohol on single human red blood cells using NIR LTRS. The significant decrease of I752 over the period of 25 minutes after the RBCs are diluted in 20% alcohol solution is probably related to the degradation of the RBCs resulting from the hemoglobin-alcohol interaction. It is also verified by the fact that the decrease of I752 become more rapid and more prominent as the alcohol concentration increases. When the alcohol concentration goes higher than 30%, the RBCs break down within 3 minutes and the cell structures collapse. The second part concerns distinguishing rectum cancer cells using Raman spectroscopy at single cell level. We are able to correctly predict 17 of 20 cancer cells and 19 of 20 normal cells based on the spectra differences between the normal rectal cell and rectum cancer cell. Thus, the sensitivity and specificity of this classification is 85% and 95% respectively. Chapter 5 presents the experiment, done during studying for doctor degree, named as measuring transmittance in rubidium vapour cells using an extremely weak coherent field of light.
