染色质调控蛋白CTCF在Ⅰ型单纯疱疹病毒

中文摘要DNA锌指结合蛋白CTCF是细胞内关键的染色质高级结构调控蛋白,其功能涉及许多重要的核内过程,包括染色质绝缘,遗传印记,远距离调控染色质相互作用,V(D)J 重组,可变剪接,RNA pol II停顿和增强子功能。I型单纯疱疹病毒(HSV-1)是一种双链DNA病毒,感染上皮细胞后进入裂解感染期,引起宿主细胞核结构显著改变。然而,关于HSV-1基因组转录复制过程中的组织、调控却知之甚少。我们的研究发现CTCF能够被招募到HSV-1复制区。CTCF的招募依赖于其锌指结构域。敲低CTCF导致病毒基因转录水平下降,病毒基因拷贝数、病毒产率减少;同时,能形成复制中心的病毒变少,大多数复制区变得弥散、不能形成成熟的复制区。ChIP-seq结果显示CTCF与病毒基因组有着广泛的相互作用。病毒基因组上CTCF结合峰与polⅡ ser2、polⅡ ser5的结合峰有相关性。ChIP-qPCR结果显示敲低CTCF影响polⅡ ser2在病毒基因组上的结合。这些结果提示我们CTCF在组织病毒复制区高级结构、调控病毒基因表达中起关键作用。
英文摘要The zinc finger DNA binding factor CTCF is the key chromatin regulator implicated in essential nuclear processes including chromatin insulation, imprinting, long-range regulatory interactions, V(D)J recombination, alternative splicing, RNA pol II pausing and enhancer functions. Herpes Simplex Virus type I (HSV-1) is a DNA virus that enters productive infection in epithelial cells and dramatically reorganizes the host nucleus. However, little is known on how the HSV-1 genome is organized during its replication. Here we report that CTCF is recruited to HSV-1 replication center. The recruitment requires the zinc finger DNA binding domain of CTCF. Knock down of CTCF resulted in repression of viral gene transcription, reduced viral genome copy number and viral yield, while, at the same time, fewer HSV-1 replication centers emerge, and many centers become diffused, less defined. ChIP-seq data showed that CTCF interacted extensively with the HSV-1 genome and demarcated regions differential with polⅡ ser2 and transcription. These data suggest that CTCF plays a key role organizing the viral replication centers and regulating viral transcription.
