中文摘要 | 寻找适合脑内多种神经活性物质免疫组织化学研究的固定方案。方法 以胆碱乙酰化酶、P 物质和γ-氨基丁酸为递质合成酶、多肽及氨基酸递质的代表,以大鼠隔O斜带O基底核此三类阳性神经元的数量、染色和形态变化为依据,检测了一系列复合固定配方对此三类抗原免疫组化的影响。结果 经0. 2 %戊二醛、4 %多聚甲醛和 0. 2 %苦味酸灌注固定和4 %多聚甲醛后固定,并经1 %硼氢化钠处理的脑组织,能很好地显示该三类神经元。此结 果还得到猫脑免疫电镜研究的支持。结论 有可能设计一种满足脑内尽可能多种类递质及合成酶免疫组化光、电 镜研究所需的复合固定配方,以期在同一实验动物获得最大程度的化学神经解剖学信息。 |
英文摘要 | To search for a suitable fixation for the simultaneous immunohistochemistry of transmitter 2 syn 2 thesizing enzymes ,neuropeptides ,and amino acid transmitters in the brain. Methods A series of compound fixation programs ,including perfusion 2 fixation ,postfixation ,and sodium borohydride treatment were designed , and their effects were evaluated on the immunohistochemistry of choline acetyltransferase (ChAT) ,substance P (SP) ,and γ 2 aminobutyric acid ( GABA) representing the three kinds of antigens in question. For each of the compound fixation groups ,the ChAT Ο、 SP 2 and GABA 2 like immunoreactive ( Ο LI) neurons were investigated in the medial septal nucleus ,the nuclei of the diagonal band ,and the nucleus basalis of Meynert in the adjacent brain sections of the rat ,respectively. And the changes in the number ,staining ,and shape of the perikarya and processes of each kind of the positive neurons ,and in the non 2 specific staining were compared among the fixation groups. Results The perfect fixation was achieved in one group perfusion 2 fixed with a mixture containing 0. 2 % glutaraldehyde ,4 % paraformadehyde ,and 0. 2 % picric acid ,postfixed with 4 % paraformaldehyde ,and with subsequent 1 % sodium borohydride treatment of the sections ,in which all the three kinds of the positive neurons were well observed. Thisfixation wasfurther supported by the immunoelectron microscopic study of the SP Ο LI Purkinje cells in the paraflocculus of the cerebellum in the cats. Conclusions Seeing the three kinds of antigens stand for most of the neuroactive substances in the brain ,we suggest that it is practical to prepare a compound fixation program satisfying immunohistochemistry of as many kinds of neurotransmitters and their synthesizing enzymes as possible to maximize information from a single experimental brain. |
