Quantitative Dynamics-Activity Relationship: Dynamics of Intrinsically Disordered Regions in the Notch 1 Transcription Complex Mediate Gene Transcription

英文摘要Transcription complexes are comprised of a DNA-bound transcription factor and coactivator proteins that are held together by cooperative and allosteric interactions. Understanding the mechanisms of action of transcription complexes continues to be a great challenge in chemical biology. The DNA-CSL-ICN-MAML1 Notch1 transcription complex (NTC) composes of a DNA-bound transcription factor (CSL: CBF1, Suppressor of Hairless, Lag-1), an intracellular domain of Notch1 receptor (ICN) and a mastermind-like family coactivator protein (MAML1). Mutation or aberrant regulation of the transcription complexes are associated with human diseases including congenital disorders, cardiovascular diseases, and cancers. As a proof-of-concept study, we performed a quantitative dynamics-activity relationship (QDAR) to analyze the dynamic properties of the IDRs in the NTC in relation to the transcriptional activity. Our study revealed a log-linear and anti-correlated relationship between the dynamic 批roperties of intrinsically disordered regions (IDRs) in the Notch1 transcription complex (NTC) and the transcriptional activity. We concluded that rigidified IRDs mediate binding of cofactors and dimerization, and leading to transcriptional activation.
