Electrical recording with iridium oxide modified microelectrodes in Drosophila melanogaster

英文摘要 | Objective Drosophila is now emerging as a favorable animal model for the studies of neuroscience.However,it remains a great challenge to employ electrophysiological approaches in Drosophila to explore the temporally precise and behaviorally relevant interactions of neurons in specific circuits,partially due to the small size of the Drosophila brain.Small,sensitive multi-unit microelectrodes for action potential and local field potential(LFP)recordings are greatly desired.Methods We developed multi-channel microelectrodes modified with iridium oxide(IrO_2) under a highly controllable anodically deposition procedure to improve their electroelectrical properties.Results Comparing to the unmodified nickel-chromium(NiCr) microelectrodes,the IrO_2 microelectrodes exhibited significantly higher capacitance and lower electrochemical impedance at 1 kHz.Recording with the IrO_2 microelectrodes to detect spontaneous action potentials and LFPs in the brains of Drosophila demonstrated significantly lower root mean square(RMS) noise,and improved signal noise ratio(SNR) and LFP sensitivity.Conclusion The results indicate that the fabricated multi-channel microelectrodes will be a useful tool for the electrophysiology-based researches especially in Drosophila and other small animals. |
