Indocyanine green loaded lipid-polymer nanoparticles for cancer imaging and therapy

英文摘要 | Indocyanine green (ICG) is an FDA-approved near-infrared (NIR) dye and has been applied in optically mediated diagnostic and photothermal/photodynamic therapy. However, unstable properties in aqueous media and quick clearance from the body limit the application of the ICG. To overcome these limitations, we report the development of ICG loaded poly(lactic-co-glycolic acid) (PLGA)-lecithin-polyethylene glycol (PEG) nanoparticles (INPs) with 39 nm, 68 nm and 116 nm. NIR imaging in vivo and photothermal therapy illustrated that 68 nm INPs showed the strongest efficiency to suppress BxPC-3 xenograft tumor growth due to abundant accumulation in tumor by the passive-targeting of nanoparticles.1 Moreover, INPs with folic acid (FA) as targeting ligand (FAINPs) were designed. FA-INPs showed excellent optical properties compared to the free ICG and exhibited highly selective localization to folate receptor over-expressing MCF-7 xenograft tumor and prolonged ICG circulation in vivo. We also fabricated doxorubicin (DOX) and ICG loaded PLGA-lecithin-PEG nanoparticles (DINPs). In comparison with chemo or photothermal therapy alone, the combined therapy of DINPs with laser irradiation synergistically induced the apoptosis and death of DOX-sensitive MCF-7 and DOX-resistant MCF-7/ADR cells, and suppressed MCF-7 and MCF-7/ADR tumor growth in vivo. Notably, no tumor recurrence was observed after only a single dose of DINPs with laser irradiation. |
