Smart Methylene Blue Nanoprobe for Imaging-Guided Cancer Photodynamic Therapy

英文摘要Photodynamic therapy (PDT) is a clinically approved therapeutic modality for the treatment various diseases including cancers. In the present study, we designed a reduction-responsive nanoplatform (BSA-MB NPs) for PDT. Our study indicated that BSA-MB NPs efficiently induced ROS. Meanwhile, BSA-MB NPs had a high accumulation with tumor-to-normal tissue ratio of 36.12±5.12 at 24 h, where the tumor and normal tissue were be clearly identified via MB-based in vivo near-infrared (NIR) fluorescence and photoacoustic dual-modal imaging. After an intravenous injection of BSA-MB NPs followed by photodynamic therapy (660nm, 150mW/cm2 for 30 min), the tumor was completely suppressed, no tumor recurrence and treatments-induced toxicity were observed. The results suggest that BSA-MB NPs generated by programmed assembly as smart theranostic nanoplatforms are highly potential for imaging-guided cancer PDT. We believe that the unique treatment modality could further the therapeutic efficiency of PDT and provide novel opportunities in tumor therapy field.
