Chemerin secreted from gonadal adipose tissue promotes ovarian cancer progression through the CMKLR1 signaling pathway

英文摘要 | Ovarian cancer is one of the most diagnosed types of cancer in women. Studies have indicated that obesity and chemokines play key roles in ovarian cancer, but whether chemerin secreted by gonadal white adipose tissue (gWAT) affects the progress of ovarian cancer through the activation of one of its receptors – CMKLR1 – remains unknown. To investigate this question, we characterized the effects of suppressing CMKLR1 function on cancer-related properties and gene expression in the ovarian cancer cell line SKOV3 by several assays, including measurement of cell proliferation, invasion, colony formation, and tumorigenicity in mice, and western blotting and Q-PCR to quantify gene and protein expression. CMKLR1 expression was knocked down using short hairpin RNA silencing, and CMKLR1 protein function was blocked using the CMKLR1 antagonist α-NETA. CMKLR1 gene silencing attenuated the expression of MMP-2, uPA, MUC16, VEGF, and MMP9 and decreased cell migration and invasion, while chemerin had the opposite effects. These results were confirmed by results obtained in vivo. We also found that both α-NETA and CMKLR1 shRNA suppressed tumorigenesis in mice and that the tumors tended to migrate to regions of higher chemerin concentration. Our data show that CMKLR1 silencing leads to the inhibition of proliferation, migration, and invasion by SKOV3 cells, whereas chemerin reverses this inhibition. The data also suggest that the migration of tumors that form in the mouse xenograft model depends on the CMKLR1 pathway. To our knowledge, this is the first report on a role for CMKLR1 in ovarian cancer formation and invasion. |
