The effect of CMKLR1 signaling on white and brown adipose tissue adiposity induced by androgen

英文摘要Excess androgen induced obesity is widely recognized as an important public health problem, whose prevalence has increased substantially in the recent decades. Chemokine-like receptor 1 (CMKLR1), a receptor of novel adipokinechemerinsecreted by adipose tissue, is associated with adipocyte differentiation, adipose tissue development, and obesity. However, the effect of chemerin/CMKLR1 on adiposity in vivo remains unclear, especially in androgen-mediated obesity. In present study, we report a critical role for CMKLR1 signaling in regulating androgen-dependent adiposity in mice white and brown adipose tissue. We constructed dihydrotestosterone(DHT) induction hyperandrogenism model in female mice and orchidectomized mediated hypoandrogenism model in male mice, respectively. In vivo androgen excess of females results in larger adipocyte size, while androgen deprivation of males induces smaller adipocyte size.Interestinly, these two effects relating to androgen are both attenuated by CMKLR1 deficiency. Moreover, wortmannin inhibition attenuated the induction effects of DHT on phosphorylated ERK in brown adipose tissue of CMKLR1 null mice, but not in WT mice, implicating the PI3K/ERK signaling pathway in the CMKLR1-dependent response to DHT. In conclusion, CMKLR1 gene deletion attenuates the adiposity effects of androgens on adipose tissue, and this effect may be mediated by PI3K/ERK signaling. These data provide evidence that CMKLR1 might be a prevention or treatment target forexcessandrogen inducedobesity. Keywords: adiposity; androgen; CMKLR1; adipokine; adipose tissue; obesity
