CMKLR1 Deficiency Leads to Testosterone-induced Bone Loss through the Regulation of Osteoblast and Osteoclast Formation

英文摘要The adipokine chemerin and its receptor, chemokine-like receptor 1 (CMKLR1), are associated with osteoblastogenic differentiation of mesenchymal stem cells (MSCs) and osteoclastogenic differentiation of osteoclast precursors in vitro. However, the effect of chemerin and CMKLR1 on bone mineral density (BMD) in vivo remains unknown. Here, we report a critical role for CMKLR1 in regulating androgen-dependent bone maintenance in male mice. We found that CMKLR1–/– males, but not females, exhibited trabecular bone loss and lower serum testosterone levels relative to wild type (WT) control mice. Furthermore, we show that male CMKLR1–/– mice had reduced mRNA levels for osteogenic markers, increased mRNA levels for osteoclast markers in bone, and decreased expression of mRNAs for testosterone synthesis enzymes in testis in vivo. In vitro differentiation models indicated that CMKLR1-deficiency caused a shift from osteogenic to adipogenic differentiation in MSCs and enhanced osteoclast formation. In addition, female CMKLR1–/– mice did not show trabecular bone mass increase which is a typical response to supraphysiological DHT treatment in vivo. These results support a model in which CMKLR1 positively regulates bone metabolism through control of testosterone production and the balance between osteoblast and osteoclast formation.
