Determination of neurotransmitters in rat telencephalon using pre-column fluorescence derivatization with high performance liquid chromatography/mass spectrometry

英文摘要 | a sensitive method has been developed for the simultaneous determination of 5 neurotransmitters glutamic acid, gamma-amino-butyric acid, dopamine, 5-hydroxytryptamine, 5-hydroxyindole acetic acid) with i, 2-benzo-3, 4-dihydrocarbazole-9-ethyl chloroformate (bceoc) as derivatization reagent on a reversed-phase hypersil bds c-18 column (100 mm x 4.6 mm, 5 mu m) with a gradient elution followed by high performance liquid chromatography (hplc) via fluorescence detection at 390 nm (excitation wavelength 333 nm) and online mass spectrometric identification. the linear range of 5 neurotransmitters was 24 fmol - 200 pmol, all the correlation coefficients were > 0.9997, the detection limit was 4.0 - 12.6 fmol (at a signal to noise ratio of 3: 1). the ms identification of 5 neurotransmitter derivatives was carried out by post-column online mass spectrometry with electrospray ion source, the ms/ms fragment ions of them were listed, and the ms/ms cleavage mode of representative 5-hydroxytryptamine derivative was analyzed. under all the above optimum experimental conditions, the contents of 5 neurotransmitters in telencephalon of 64 rat (classified as 2 groups, a group: quiet contrast group, b group: training group) at 4 states (a: quiet state, b: 1 h after exercise, c: at exercising exhaust, d: 12 h after exercising exhaust) were determined. it is indicated that a long period of time medium intensity training can effectively improve mice exercise capacity and obviously promote to restore central fatigue after resistance exercise. the established method exhibits high sensitivity and excellent reproducibility, and provides a new technology for the simultaneous determination of monoamine and amino acid neurotransmitters in rat telencephalon. |
