Effect of High-fat Diet on Genome-wide Profiles in Ovary and White Adipose Tissue of Immature Female Mice

英文摘要 | Obesity, a chronic multifactorial disease with high prevalent in many areas of the world and displays excess fat accumulation, is a main source of morbidity and mortality. Presently, overweight or obese increases the relative risk of metabolic diseases in women including diabetes, low and sterility which causes lots of attention these years. By now, The mechanisms underlying these effects, especially in young women, is largely unknown. To explore these mechanisms, we established a high-fat diet (HFD) model of obesity in immature female mice. Microarray was performed to characterize the effects of HFD on reproductive development, and to explore correlation between reproductive system and metabolism. In ovaries of the HFD group, there were 208 differently expressed mRNAs>1.3 fold change, including 98 upregulated and 110 downregulated and 295 differently expressed lncRNAs >1.3 fold change, including 63 upregulated and 232 downregulated. In white adipose tissue, there were 625 differently expressed mRNAs>1.3 fold change, including 220 upregulated and 605 downregulated, and 1595 differently expressed lncRNAs>1.3 fold change, including 1320 upregulated and 275 downregulated showed significantly different expression profile in HFD groups, respectively. Our results reveal significant differences between the transcriptomes of the HFD and control groups in both ovaries and white adipose tissue that provide clues to the molecular mechanisms of diet-induced female reproductive dysfunction and metabolic disorders, as well as biomarkers of risk for these disorders. |
