Rapid isolation of placenta extracellular vesicles via affinity chromatography

英文摘要Extracellular vesicles (EVs) can mediate intercellular communication by transferring cargo proteins and nucleic acids between cells. Different biophysical and biochemical properties can be used to isolate EVs, including size, mass density, shape, charge, and antigen exposure. But, rapid isolation of specific organs EVs, particularly for placenta EVs, from plasma is still a big challenge. Our preliminary researches have demonstrated that the chondroitin sulfate A presented on the surface of trophoblast cells and some EVs secreted by trophoblast cells. The protein rVAR2 can specifically bind to trophoblast cells expressing the chondroitin sulfate A. In this study, we used the rVAR2 protein as a substrate to separate extracellular vesicles in plasma by affinity chromatography. Then we examined the morphology, number, and purity of isolated EVs by TEM and Elisa. Our results show that, after home-made affinity chromatography, a certain amount of exosomes which expressing CD63 marker can be isolated. At the same time, we also developed the new technique for EVs immune localization and analyzed the nucleic acids and proteins in trophoblast cells to ensure isolated EVs and trophoblast cells have ‘homology’. More study need to be performed in order to improve the accuracy and efficiency isolation of placental EVs and to provide an novel noninvasive diagnosis method for placental diseases research.
