Fluorescent/Magnetic Nanoprobes of High Specificity for Detection of Triple Negative Breast Cancer

英文摘要Triple negative breast cancer, one of the most malignant cancers known to date, has a high degree of malignancy with poor prognosis. However, there is no efficient method available clinically to detect the triple negative breast cancer and, therefore, a new and/or innovative method for an early stage detection of this cancer is warranted. Recently, upconversion of rare earth nanomaterials with equal size, narrow spectrum, long luminous life and stable performance has piqued the curiosity of scientific community. In this work, magnetic and fluorescent upconversion nanoparticles (UCNPs) were synthesized via high temperature pyrolysis method and then modified with polyacrylic acid (PAA) and GE11 peptide. The resulting nanoprobe demonstrated a high performance with good affinity and specificity to triple negative breast cancer expressing high level of epidermal growth factor receptor (EGFR). In addition, the freshly synthesized UCNPs exhibited a good dispersibility and photostability after modification with PAA. Under optimized experimental conditions, the newly developed UCNPs exhibited satisfying performances for the detection of the triple negative breast cancer. In vitro test revealed that the detection limit in culture solution is less than or equal to 100 cells/mL indicating that the newly developed nanoprobe is a promising candidate for detection of the triple negative breast cancer with extraordinary capability to enhance the magnetic resonance (MR) and fluorescence images.
