IDENTIFICATION OF THE OROTIDINE-5’-MONOPHOSPHATE DECARBOXYLASE GENE OF THE OLEAGINOUS YEAST RHODOSPORIDIUM TORULOIDES

中文摘要 | Rhodosporidium toruloides is an excellent microbial lipid producer of great industrial potential [1]; yet, there is no effective genetic tool for rational engineering this microorganism. Orotidine-5’-monophosphate decarboxylase (OMPdecase), usually encoded by the gene ura3, and has been widely employed as a genetic selection maker in Saccharomyces cerevisiae [2], other yeasts [3] and fungi. To develop a marker recycling system for R. toruloides, the ura3 gene of R. toruloides (Rt-ura3) was isolated using methods of degenerate PCR together with RACE. Results showed that the cDNA and genomic sequence for Rt-ura3 were 840 bp and 1017 bp, respectively, indication that the gene contained 4 extrons and 3 introns. The encoded polypeptide held an amino acid sequence of 279 residues with significant homology to those of OMPdecases from other yeasts. A shuttle vector pYES2/CT-Rt-ura3 was constructed via site-specific insertion of Rt-ura3 into the commercial vector pYES2/CT. Transformation of the vector into S. cerevisiae BY4741, a URA3-deficient yeast strain, ensured the viability of the strain on synthetic dextrose agar plate without uracil, suggesting that the isolated Rt-ura3 was functionally equivalent to the ura3 gene from S. cerevisiae. Further work is going on in this laboratory to engineer oleaginous yeasts based on this information. |
