Separation of Chitooligosaccharides (DP2-6) Monomers and the Potent Angiogenic Inhibition Effect of Chitohexaose in vitro

中文摘要chitooligosaccharides(dp2-7, 95% deacetylated) were separated to five monomers for the first time by a acid-ion-exchange resin with linear gradient of hcl solution from 2.0m to 3.0m, and was followed by a charcoal column to remove hcl from sugar. hplc and maldi-tof mass spectrum were performed to analysis the monomers. chitobiose was 341da with retention time of 10.2min. chitotriose was 502da with retention time of 9.6min. chitotetraose was 663da with retention time of 8.6min. chitopentaose was 824da with retention time of 7.3min. chitohexaose was 985da with retention time of 6.9min. the molecular masses were exactly without any acetyls. in our previous study, we found the anti-angiogenic activities of cos and in present research, the cos monomers were used to confirm the exact component which had the potent anti-angiogenic activity. the results showed chitohexaose had no toxicity to normal huvecs, but could inhibit the proliferation and migration of induced huvecs in a dose-dependant manner.
