Fundamental structure and properties of alginate/chitosan hydrogel microcapsule surface and its interaction with protein/cells

| 中文摘要 | alginate/chitosan (aca) hydrogel microcapsules have been widely studied as devices for immuno-isolation and transplantation of living cells for its good biocompatibility. the fundamental structure and properties of microcapsule surface is one of the most important factors that determine the protein adsorption, cellular behavior and fibrotic overgrowth (in vivo biocompatibility). in the present work, surface morphology of aca microcapsules was analyzed firstly using a scanning electron microscopy and a noncontact, three-dimensional optical interferometer. the chemical composition of the aca microcapsule membranes was then determined using x-ray photoelectron spectroscopy (xps). the surface wettability and charge were determined by contact angle and zeta potential measurements respectively. after that, the effects of surface properties on bovine fibrinogen (fgn) adsorption onto aca microcapsules and its in vivo biocompatibility were evaluated. the results showed that the surface of aca microcapsule exhibited a granular structure and the surface roughness could be controlled easily by changing complex composition. xps results suggested that both alginate and chitosan were present within the outer 100 å of the membranes. there was net negative charge on the surface of aca microcapsules which varied with the complex composition. the increase in both surface roughness and zeta potential resulted in an increase in the amount of fgn adsorbed. moreover, the higher zeta potential was, the stronger the protein-surface interaction between fibrinogen and aca microcapsules was. more protein molecules adsorbed spread and had a greater conformational change on rougher surfaces for more surfaces being available for protein to attach. as a result, aca microcapsules with rougher surface and higher zeta potential that had been implanted into the peritoneal cavities of male mice were found to elicit a more severe cell adhesion and fibrotic overgrowth. |
