A Plasma pharmacology strategy for discovering multi-components interactions of Xian-Ling-Gu-Bao capsule on exerting anti-osteoporosis activity

英文摘要 | Introduction: The famous traditional Chinese medicine prescription Xian-Ling-Gu-Bao(XLGB) capsule has been proven to be clinical efficacy on the treatment of osteoporosis in China. However, the mechanism of absorbed components in XLGB after oral administrated is too complex to ascertain in a traditional way. As many investigators hypothesized that Chinese medicine may work by integrative effects of multi-components in plasma rather than single effective constituents, a holistic strategy is urgent to be proposed in order to investigate the relationship among the absorbed components in blood. Objectives: The present study aims to establish a method for the preparation of drug-containing plasma, whereby the interaction effects of absorbed components on exerting anti-osteoporosis activity and their underlying mechanism could be elucidated in a simplified way. Materials and Methods: Rat plasma after oral administration of XLGB capsule was prepared with the guidance of comprehensively evaluating osteogenic activity on MC3T3-E1 pre-osteoblasts and bone resorption activity on RANKL-induced RAW264.7 cells. The components in the XLGB-treated plasma with anti-osteoporosis activity in vitro were subsequently determined by ultra performance liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry. Representative components were selected from these components, which considering different structural types originated from individual medical herbs with detectable peak areas. A combinatorial fraction was formulated with representative compounds according to their concentrations in plasma, while further study was performed on comparing osteogenic activity of the extracted combinatorial fraction with representative components at their concentration in plasma. Finally, potential synergistic combination among representative components was predicted by NIMS, a network target computational tool.Results and Conclusions: Our present study successfully established a general method for the preparation of drug-containing plasma, which could neglect the interference of matrix in blood. According to the chemical and biological investigation on plasma, we evidenced that the therapeutic efficacies of XLGB are achieved by combinatorial components rather than single compounds. Altogether, this work provided a novel plasma-pharmacology strategy to study the complexity absorbed components in herbal medicine and could be used to guide the quality control of XLGB. |
