Novel cyanine 5 conjugated magnetic beads for rapidly and efficiently detection of OATP1B3 over-expressed circulating tumor cells

英文摘要Circulating tumor cells (CTCs) are cancer cells in peripheral blood spontaneously released by solid tumor or metastatic lesions, sometimes caused by surgery operation. Recently, CTCs detection technologies including PCR, fluorescenceactivated cell scanning/sorting (FACS), magnetic-activated cell separation (MACS) and Cell Search have been widely used in research and clinic. Specially, Cell Search is the only clinically applied CTCs capture technology via EPCAM antibody modified magnetic beads. However, some defects limited its Figure 1. Some special cyanine dyes have been reported to display excellent targeting property by transporting of OATPs which over-expressed in most tumor cells. In this work, we firstly discovered superior targeting property of Cyanine 5, a commercial fluorescent cyanine dye. Accordingly, we developed a new fluorescent diagnostic technique that combines Cyanine 5 with magnetic beads for dual separation of OATP1B3 over-expressed CTCs. This technique overcame defects of antibody and showed great potential for further application of clinical capture of CTCs
