Tumor-targeted multifunctional Core@Shell magnetic nanoprobes for near-infrared photodynamic therapy/chemotherapy of gastric cancer stem - like cells

英文摘要Developing safe and effective nanoprobes for targeted imaging and selective therapy of gastric cancer stem cells (GCSCs) has become one of the most promising anticancer strategies[1]. Herein, amphiphilic polymer modified core@shell magnetic nanoparticles (CoFe2O4@MnFe2O4-PMA=Co@Mn-PMA MNPs)-based PGylated multifunctional nanoprobes loaded with Ce6 were established with conjugated all-trans retinoic acid (ATRA) as the targeting ligand. In this study, the CoFe2O4 core could enhance magnetic anisotropy, and the MnFe2O4 shell could improve the magnetization value. Meanwhile, PG dendrimer was utilized to improve both the loading efficiency of photosensitizer Ce6 and the biocompatibility of Co@Mn-PMA MNPs. More importantly, the ATRA worked not only targeting molecules to achieve highly effective tumor-targeted drug accumulation, but also as the chemotherapy drug[2]. The application of multifunctional nanoprobes (Co@Mn-PMA-PG-ATRA@Ce6) for in vitro and in vivo therapy achieved the combined antitumor effects upon the NIR irradiation (Fig.1). In addition, we monitored the bio-distribution and excretion pathway of the nanoprobes based on the NIR fluorescence from Ce6 and MR/photoacoustic signal from Co@Mn MNPs, and the results proved that the nanoprobes had a great ability for tumor-specific drug delivery and efficient accumulation of tumor sites. What’s more, the nanoprobes inhibited tumor growth remarkably upon NIR laser irradiation, and even extended survivability of the gastric cancer-bearing mice. We therefore conclude that the Co@Mn-PMA-PG-ATRA@Ce6 holds great promise for clinical applications in anti-tumor therapy with tumor imaging or drug tracing.
