Astrocytes in the ventromedial hypothalamus mediate anxiety in mice.

英文摘要Ventromedial hypothalamus (VMH) is important in regulating metabolism and mental disorders, such as anxiety and depression. Recent evidence has demonstrated that astrocytes in hippocampus and amygdala play key roles in mediating anxiety behavior through secreting FGF-2 and other factors. Our previous study has shown that optogenetic activation promotes FGF-2 secretion from astrocytes. However, the exact role of astrocytes in VMH in regulating anxiety behavior is unclear. In present study, combining pharmacogenetic, microdialysis, brain slice electrophysiology and behavior analysis, we found that chronic stressed mice displayed typical anxiety-like behavior, GABA concentration in VMH nucleus was significantly elevated, and optogenetic activating GABAergic neurons in VMH induced significant anxiety-like behavior. Using S100-Cre mice, we further demonstrated that pharmacogenetic inhibiting VMH astrocytes through CNO reduced anxiety-like behavior significantly; More interestingly, a gender based difference in anxiety outcome is also observed: male mice show more anxiolytic phenotype than female after CNO administration. Taken together, our study emphasized VMH astrocytes role in regulating anxiety-like behavior in mice. Considering the role of astrocytes in reuptake and metabolism of neurotransmitters, more study is needed to dissect the molecular mechanism of VMH astrocytes-mediated anxiolysis.
