英文摘要Using hairpin dna probe self-structure change during dna ligation process, a sensitive, label-free and simple method of e. coli dna ligase assay via a home-built high-resolution surface plasmon resonance (spr) instrument was developed. the dna ligation process was monitored in real-time and the effects of single-base mutation on the dna ligation process were investigated. then an assay of e. coli dna ligase was completed with a lower detection limit (0.6 nm), wider concentration range and better reproducibility. moreover, the influence of quinacrine on the activity of e. coli dna ligase was also studied, which demonstrated that our method was useful for drug screening.
