英文摘要 | A novel Coanda effect electrospray ionization source (CEESI) was designed and applied in the analysis of the digests of bovine serum albumin (BSA) and proteins extracted from HeLa cells. Firstly, the effects of various organic solvents on the mass spectrometry signal of a standard peptide were investigated. With ethanol vapor, the signal of standard peptide was increased by about 1000 fold, and the sequence coverage of 1 ng of BSA was improved by about 10%. Finally, under the optimized conditions, 6, 10, 20 and 50 ng of HeLa cell tryptic digests were qualitatively analyzed with nano. HPLC. CEESI. LTQ XL MS system. In comparison with commercial nano. ESI, the number of identified peptides and proteins were increased by 20% - 360% and 20% - 200%, respectively, demonstrating that the CEESI ion source is promising in the in. depth profiling of trace proteomic samples. |
