英文摘要Recent advances in the study of triplex deoxyribonucleic acid (DNA) are reviewed from three aspects: Firstly, triplex DNA stability study. The stability of triplex DNA is not only determined by the oligonucleotides base sequence, but also dependent on several other factors, such as pH, concentration of cations, DNA intercalative agents and copolymer etc. Secondly, triplex DNA application. The formation of triple helix DNA offers-a powerful tool for genetic manipulation and genetic therapy. The triplex DNA can be used to inhibit DNA transcription and replication, generate site-specific mutations, cleave DNA, and induce homologous recombination. Thirdly, triplex DNA detection. Many analytical methods have been developed for the sensitive detection of triplex DNA, such as UV-visible absorption spectrum, fluorescence analysis, atomic force microscopy and electron paramagnetic resonance spectra etc. In the end, the potential development of triplex DNA is addressed.
