英文摘要 | A new synthetic support, monosized, non-porous, cross-linked bead CL-poly(glycidyl methacrylate) (CL-PGMA) containing active epoxide groups was synthesized and used as the support of affinity chromatography. The concentration of the epoxide groups on the surface of CL-PGMA beads was measured and the non-specific adsorption to protein was characterized. Trypsin inhibitor was chosen as the ligand. The trypsin was purified 2.5 times by using the trypsin inhibitor immobilized CL-PGMA affinity column. The chromatographic characterization was shown that the CL-PGMA bead was very suitable for the high performance affinity chromatography. The effective elimination of non-specific adsorption was carried out by adding 1% acetonitrile in mobile phase. The recovery of human serum albumin was more than 99%. |
