Separation of four basic proteins in the mixture by capillary electrophoresis with a new chemical modification column

英文摘要 | Chemical modification of internal wall of capillary by in situ polymerization method using a reagent containing two function groups, methacryloxypropyl trimethyoxysilane (KH-570) as coupling agent and glycidyl methacrylate (GMA) as coating material in the presence of initiator was carried out. After hydrolysis of polyglycidyl methacrylate (PGMA), the novel modified capillary column was prepared. The electrophoretic evaluation of the modified column was performed. The results showed that electroosmotic flow (EOF) was largely reduced and the modified column can be applied to the basic proteins possessing serious adsorption property. In the 1 mM phosphate buffer (pH 5.0), the efficiency of the column for lysozyme, cytochrome C, ribonuclease A and alpha-chymotrypsinogen A in their mixture were relative high with 2.8, 4.6, 2.8 and 3.5 x 10(5) theoretical plates per meter. and RSD (n=4) expressed as migration time was relative low i.e., 1.2, 0.71, 0.60 and 1.1 (%), respectively. The column may be used in a wide pH range (pH 3-7). The above four basic proteins in the mixture have been successfully separated and the column possesses good stability. |
