A comparative study of bacterial community structures in the sediments from brominated flame retardants contaminated river and non-contaminated reservoir

英文摘要 | The bacterial community structure in sediments was studied by rapid amplified ribosomal DNA restriction analysis (ARDRA) combined with phylogenetic analysis of 16S rDNA clone libraries. Comparative analyses were carried out on two sediment samples collected from a non-contaminated reservoir, and a brominated flame retardants contaminated Lian River, and two sediment samples enriched with decabromodiphenyl oxide for one and two months. Based on the bacterial diversity, sediments from the reservoir and Lian River contained a more diversified bacterial population compared to those after enrichment. The composition of bacterial strains, the dominant strains, and the number of gene clones identified to each family of bacteria which significantly differed between sediments from the contaminated river and non-contaminated reservoir. From each of the two types of sediments, 100 rDNA clones had matched sequences in the GenBank databases, 91 clones were in the unculturable bacteria category, and 43 clones possessed less than 95% sequence similarity. In the two enriched samples, the bacterial population structure became less diversified. 28 out of 82 sequences matched with sequences of unculturable origin with 17 clones sharing less than 95% homology with known sequence deposited at the GenBank. Compared to the original river sediments, the bacterial community composition was altered after the enrichment. The enriched samples contained fewer phyla, but more abundant within-phyla members of bacteria. The dominant strains are in the classes of a-and beta proteobacteria, and the ratio of culturable bacteria was also increased. Results from this study indicate that different contaminants can cause alteration in the bacterial community structure. |
