用nrDNA的ITS序列探讨柽柳科部分属、种的系统学位置

中文摘要本文主要对来自怪柳属(TamarixL)、水柏枝属(Myricaria Desv.)、琵琶柴属(Reaumurial L)的21个种采用PcR直接测序的方法,对核糖体DNA中的内转录间隔区(ITS)序列及5.8SrDNA的3端序列进行测定对,所获得的608个碱基序列位点进行全面分析。测得21种植物的ITS1和ITS2的长度、分析序列中GC含量、应用系统软件分析基因序列间的绝对遗传距离和相对遗传距离、用分枝界限法(Bound and Branch)寻找作最简约系统树。结果表明:开S一1片段的长度在254 bp-269bp之间,rrs-2片段的长度在225bp-253bp之间ITS在怪柳属与水柏枝属间存在10bp~19bP的长度差异,在怪柳属与红砂属间存在24bp-28 bp的长度差异。各属内不同类群的仃S片断长度差异不大5.8SrDNA的3端近83个碱基序列在属内和属间无长度差异。怪柳属内ITS一1区的序列长度为259 bp~260bp,开s一2区的序列长度为244 bp一247bP,G+c含量在rTs一1区的变化范围为61%一63%,在开S一2区为58%一65%。水柏枝属内rTS一1区的序列长度为264 bp~269bp,rrs一2区的序列长度为246饰一253bp,G+c含量在仃S一1区的变化范围为50%一53%,在rrs一2区为48%~53%。在不考虑外类群时,排序后的序列(包括gaP在内有180个变异位点,其中115个为信息位点。仃S一1区的变异位点有77个。ITS-2区的变异位点有93个。信息位点在ITS-1区有46个,ITS-2区有69个。运用PAUP软件进行的系统发育最简约树,其总步长为466步,一致性指数CI=0.8584,保持性指数RI=0.8622。当gaP处理为数据丢失(missing)时,得到一个同等最简约树,其中简约树的长度为400步,其一致性指数(CD)和保持性指数(RI)值分别为0.8400和0.8535。系统发育分析表明:秀丽水柏枝应从水柏枝属中分出,但又不应归人怪柳属,故应另立新属。另外,研究分析为目前分类上存有争议的短毛怪柳、白花怪柳及甘蒙怪柳p的划分提供了分子生物学证据。结果表明:短毛怪柳系统关系与刚毛怪柳很近,可能是刚毛怪柳的变异;白花怪柳也不能简单作变异处理;甘蒙怪柳不是多枝怪柳或中国怪柳的异名,是一独立种;多花怪柳应被视为独立种。The sequences of the internal transcribed spacer (ITS) and 3 ' end of 5.8SrDNA gene of 21 representative species of three genera in Tamaricaceae were systematically studied by using PCR technique and all 608 sites, the length of ITS 1 and ITS2, G+C content, absolute distance, mean distance and phylogenetic tree were analyzed by PAUP4.0 software. The result indicated the sequences of ITS-1 rang from 254bp to 269bp and 1TS-2 range from 225bp to 253bp. The sequences difference between Tamarix and Myricaria were from lObp to 19bp and between Tamarhc and Reaumuria were from 24bp to 28bp. Meanwhile, sequences among three genera had a little difference and 3 ' end of 5.8SrRNA in 83bp had no difference within or between genera. Sequences of ITS-1 in Tamarix rang from 259bp to 260bp and ITS-2 range from 244bp to 247bp and percent G+C content of ITS1 rang from 61 to 63 and ITS2 rang from 58 to 65.Sequences of ITS-1 in Myricaria rang from 264bp to 269bp and ITS-2 range from 246bp to 253bp and percent G+C content of ITS1 rang from 50 to 53 and 1TS2 rang from 48 to 53.The aligned length of ITS region was 608 bp, which provided 180 variable sites and 115 phylogenetic informative sites when gaps were treated as missing. ITS-1 region provided 77 variable sites and 46 phylogenetic informative sites.ITS-2 region provided 93 variable sites and 69 phylogenetical informative sites. PAUP parsimonious tree showed that tree length is466,CI=0.8584 and RI=0.8622. Parsimonious tress, with length of 400,consistency index (CI) of 0.8400 and retention index (RI) of 0.8538 were obtained by Bound and Branch analysis. Sequences demonstrated that Myricaria elegans, which differed from Tamarix and Myricaria, should not be included in Tamarix L.but a new genus.The study gave molecular evidence to taxonomy of T. albiflonum, T. karelinii and T. austromongolica which were disputed. The results indicated that T. karelinii, which was close to T. hispida in taxonomy, was not a natural species but a variation. T. albiflonum was not considered simply as a variation. T. austromongolica was not homonymous as T. hohenackeri or T. chinensis but a natural species. T. hohenackeri was considered a natural species.
