巨穗小麦种质的细胞遗传学研究

中文摘要本论文进行了巨穗小麦种质材料培育中主要外源基因供体亲本八倍体小偃麦中3(多年生1号)的染色体组型分析、部分巨穗种质的C-分带分析和高分子量谷蛋白(HMW-Glu)的分析、对一巨穗种质材料特导性状的基因单体系定位。结果表明:1. 八倍体小偃麦中3中附加的中间偃麦草的一组染色体C-分带特征有别于普通小麦的21条染色体,得到附加的一组中间偃麦草染色体C-分带带型。2. 对5个不同类型巨穗小麦种质材料进行C-分带分析,发现均有一对特征染色体存在,初步确定这对染色体为小麦-黑麦1BL/1RS易位染色体。3. 对9个巨穗小麦种质材料进行高分子量谷蛋白(HMW-Glu)电泳分析,确定了每一材料高分子量谷蛋白亚基组成,发现Glu-B1控制的7+9亚基在材料中普遍存在,由Glu-A1制的亚基大部分材料缺失,Glu-D1控制的亚基大多数材料为2+12亚基,少数材料中为5+10优质亚基。4. 对于巨穗小麦种质材料95-211进行了初步基因定位,结果表明,对于株高性状的检测,1A、2A、7A、1B、3B、3D单体系显著或极显著地低于二体系,其中1A、2A、7A、1B、3D的效应相对明显。对于穗长性状的测验,3A、5A、3B、4B单体系穗长显著或极显著高于二体系,其中5A的效应最强。对于小穗数性状的检测,1B、3B、6B单体系小穗数显著高于二体系。
英文摘要The octaploid wheat-Agropyrum intermedium Zhong 3 (2n=56) and some giant wheat germplasms were studied by chromosome C - banding in this paper. Electrophoretic analyses of the High-molecular-weight subunits of wheat Glutenin were carried out in some giant spike wheat germplasms. Localization of genes for particular traits of "95-211" was carried out by using "Abbondanza" monosomic lines. The results are as follows: 1. Seven pairs of Ag. intermedium chromosomes in Zhong 3 were detected to be different to the chromosomes of common wheat. The C-band of additional Ag. intermedium chromosomes was obtained. 2. 5 giant spike wheat germplasms were studied by C-banding. A identical chromosome is existed in each, and it is primarily recognized as wheat-secale 1BL/1RS translocation. 3. The High-molecular-weight subunits of wheat Glutenin in 9 giant spike wheat germplasms were determined by electrophoretic analysis. The subunits 7+9 controlled by Glu-B1 exist almost in each germplasm. The subunits controlled by Glu-A1 are Null in most of germplasms. The subunits controlled by Glu-D1 are 2|12 in most of them, and 5+10 in some other germplasms. 4. Preliminary monosomic analysis of "95-211" was carried out by using the monosomic lines "Abbondanza". The chromosomes 1A、2A、7A、1B、3B、and 3D carry genes which control the plant height. The chromosomes 3A、5A、3B、4B carry genes which control the spike length, and 5A might carry major effect genes. The chromosomes 1B、 3B、 6B carry genes which control the spikelet number.
