中文摘要the complexity of proteome brings great challenge and opportunity to promote the development of new platforms for liquid phase based proteome analysis. in our recent study, to achieve high throughput, high accuracy and high sensitivity proteome analysis, 2d-sec-micro rplc for protein separation, a hollow fiber based interface to remove acetonitrile in the eluants of microrplc and adjust the ph to 8.0, immobilized trypsin reactor for on-line protein digestion, and microrplc-esi-ms/ms for peptides separation and identification were integrated, which could be fully automatically controlled with a self-designed software. by such an integrated platform, within 21 hr, 2014 proteins were identified from rat brain with fdr<1%. compared to the widely applied bottom up strategy, not only the analysis throughput could be obviously improved by integrating on-line protein digestion, but also the accuracy for protein identification could be increased by protein fractionation with 2d-hplc.
