Metabolomic identification of potential biomarkers for acute graft rejection of renal transplantation by using RPLC and HILIC coupled with mass spectrometry

中文摘要acute graft rejection is a major post-transplant complications of renal transplantation, the success of renal transplantation is closely associated with the ability to control rejection. therefore, early detection and accurate diagnosis are critical in the management of patients receiving renal transplantation. metabonomics is the identification and measurement of metabolic profile dynamics of living systems to toxin or drug, environmental changes and diseases. as a powerful analytical platform, the application of metabonomics has increased in therapy and novel potential biomarker discovery. in our study, a metabonomics technique based on ultra performance liquid chromatography (uplc) coupled with q-tof mass spectrometry was employed to investigate plasma metabolic profile of acute renal graft rejection patients in order to find potential disease biomarkers and to reveal its pathophysiological changes. the reversed-phase chromatography and hydrophilic interaction chromatography were combined to achieve more comprehensive metabolism. creanitine and indoxyl sulfate which are clinical markers of renal injury were also found as potential discriminating metabolites in our study. moreover, phospholipids- metabolism and ffa- metabolism alterations were identified in acute graft rejection of renal transplantation patients. these results suggest that combined lc-ms metabonomic approach is applicable to investigate the metabolic abnormality in the acute graft rejection of renal transplantation rats. these potential biomarkers may be useful molecules for the clinic in either diagnosis or therapy of acute graft rejection.
