Orthotopic xenograft of Wilms' tumor in nude mice:Biological characteristics of the models

英文摘要Objective To establish orthotopic xenograft models of human Wilms' tumor in nude mice. Methods Histologically intacted human Wilms' tumor fragments from one female baby ( stage II ) in 14 months of age were transplanted into subrenal capsular space of nude mice. Subsequent generations of transplantations were carried out in similar fashion. The first, second, third and fourth generations of tumor transplants numbered 2, 5, 5 and 18 respectively. The models were removed and weighed after 8 weeks. The first, second and third generations of mice were sacrificed after 8 weeks. Every 3 nude mice out of fourth generation were sacrificed weekly from week 3 to 8 to study tumor growth dynamics. The cell cycle distribution, apoptosis and ploidy of the original tumor and the transplants were studied with flow cy-tometry. Tumors were examined for morphology and immuno-stained for intermediate filaments, CK18, vimentin and desmin. PCR amplification of three microsatellites D14S68, D18S69 and, D20S199 in genome were performed to clarify its human origin. Results Twenty-seven out of 30 orthotopic xenografts of human Wilm's tumor were detected in nude mice after four generations. Its growth curve was exponential. DNA index and cell cycle/apoptosis distributions were similar in each generation. All transplanted tumors retained the original histological and immunohistochernical staining characters. Furthermore, mi-crosatellite DNA sequences in genome of transplanted tumors were identical with that of original human Wilms' tumor. Conclusions The orthotopic xenograft model of human Wilms' tumor is a close reproduction of the original tumor. It serves as a valuable tool in the biological and therapeutic study of human Wilms' tumor.
