Paeoniflorin attenuates the impairment of learning behavior and hippocampal neuron apoptosis caused by chronic cerebral hypoperfusion in rats

英文摘要 | Paeoniflorin (PF), a major constituent of peony root (Paeonia lactiflora PALLAS), was proved to be neuroprotective in middle cerebral artery occlusion model. Because peony root has long been used as a component of traditional Chinese prescriptions to treat certain types of dementia, we investigated whether PF attenuates cognitive damage in rats with chronic cerebral hypoperfusion. PF (1.25 and 2.5 mg/kg) were administered s.c. after surgery. Sham-operated animals served as control for the surgery. Eight weeks after the onset of cerebral hypoperfusion, the rats were tested in a hippocampus-related learning paradigm, the Morris water maze. PF (2.5 mg/kg) restored cerebral hypoperfusion-related learning dysfunction and prevented CA1 neuron loss. Cerebral hypoperfusion led to activate glia cell and promote the expressions of nucler factor-kappa B (NF-κB), and PF could modify these changes. We conclude that PF attenuates cognitive deficit and neuron damage by suppressing glia cell activation and the expression of NF-kB in glia cells. |
