| 英文摘要 | A more practical and efficient synthetic route of laninamivir octanoate,a new drug for anti-flu treatment,was established from N-acetylneuraminic acid via esterification,acylation,cyclization,azide substitution,deprotection,protection,methylation,and hydrolysis follow ed by reaction w ith N,N-bis(tert-butoxycarbonyl)-1-guanylpyrazole,deprotection via sodium hydroxide,protection via diphenyldiazomethane,esterification with acyl chloride and deprotection via trifluoroacetic acid.The overall yield of the thirteen-step synthesis was up to 16% and the structure of the target compound w as confirmed by ~1H-NMR, ~(13)C-NMR,LC-MS and HR-MS. |
