| 英文摘要 | OBJECTIVE To develop a rapid and sensitive analytical method for quantification of hydroxyethyl starch in human plasma and urine.METHODS The hydroxyethyl starch was precipitated by actone after deproteinization of the plasma and urine sample with trichloroacetic acid.After the removal of the supernatant,the hydroxyethyl starch precipitate was dissolved and hydrolyzed into monomer with trifluoroacetic acid.After drying and dissolving in phosphate buffer solution,the glucose determination was performed using an enzymatic test based on hexokinase/glucose 6-phosphatese ,and the concentration of hydroxyethyl starch was calculated according to the concentration of glucose.RESULTS The linear range of the calibration curve for hydroxyethyl starch was 0.25 - 10.0 mg · mL-1 which covered the actual plasma concentration.The intra-day and inter-day relative standard deviations of all the check samples were less than 13.2% ,and the relative error was in the range of -2.0% to -0.7%.The assay procedure has been validated in term of accuracy,precision,linearity,selectivity and robustness.150 samples could be analysed per day.CONCLUSION The method is proved to be sensitive,rapid and suitable for the determination of hydroxyethyl starch in human plasma and urine as well as the investigation of hydroxyethyl starch pharmacokinetics. |
