英文摘要Objective To investigate the selective de-O-acetylation of N-(9-fluorenylmethoxycarbonyl) -N'- (2-acetylamido-2-deoxy-beta-D-glucopyranosyl) -L-asparagine (Fmoc-Asn (Ac_3GlcNAc) -OH). Methods The Fmoc-Asn(Ac_3GlcNAc) -OH was treated in ultrasound sonication conditions for selective de-O-acetylation. The optimal conditions were obtained through the monitoring of HPLC profiles of reaction. Results Under the condition of alkali reagent, the reaction had many impurities. But under the condition of ultrasonic, the reaction was very pure and had a high rate of production. Conclusions Fmoc-Asn(Ac_3GlcNAc) -OH and Fmoc-Ser(Ac_3GlcNAc) -OH can be selectively de-O-acteylated under ultrasound sonication with more than 90% yield.
