Determination of Irbesartan in Human Plasma by Liquid Chromatography-Tandem Mass Spectrometry

| 英文摘要 | OBJECTIVE To develop a LC/MS-MS method for direct quantification of irbesartan in human plasma.METHODS An aliquot of 50 muL plasma was treated by a simple protein precipitation using acetonitrile,then separated on a Zorbax SB C_(18) column.The mobile phase consisted of acetonitrile-water-formic acid(90∶10∶0.25).A triple quadrupole tandem mass spectrometer equipped with an electrospray ionization(ESI) source was used as detector and was operated in the positive ion mode.Selected reaction monitoring(SRM) mode using the precursor to product ion of m/z 429 to m/z 207 and m/z 515 to m/z 276 was performed to quantify irbesartan and the internal standard telmisartan,respectively.RESULTS The linear calibration curves were obtained in the concentration range of 2.0~5 000 mug·L~(-1).The lower limit of quantification was 2.0 mug·L~(-1).The inter-and intra-day precision(RSD) was below 8.3%,and the accuracy(RE) was within4.1% calculated from QC samples.The method gave the mean recovery 90.9% and 90.4% to irbesartan and internal standard telmisartan,respectively.The total chromatographic run time was only 3.6 min.The method was employed in a pharmacokinetic study of irbesartan after an oral administration to 18 healthy volunteers.CONCLUSION The method was proved to be selective,sensitive,and suitable for clinical investigation of irbesartan pharmacokinetics. |
