英文摘要Oral tyrosine kinase inhibitors have been used for the treatment of cancers in recent years. Multiple tyrosine kinase inhibitors are substrates of ATP binding cassette transporter superfamily. The solubility of tyrosine kinase inhibitors is poor in gastrointestinal tract, and the efflux transporters are unlikely to be saturated. Therefore, compared with other drugs, absorption of these drugs is affected more significantly by efflux transporters. In addition, distribution and elimination of tyrosine kinase inhibitors are modulated by efflux transporters located in various tissues. In this review, how the efflux transporters modulate the disposition of seven tyrosine kinase inhibitors in vivo is summarized. The effect of genetic polymorphism of efflux transporters on the pharmacokinetics and pharmacology of these drugs is also reviewed.
