| 英文摘要 | The pulsatile controlled-release tablets were prepared with metoprolol tartrate as the model drug and the cellulose acetate-hydroxypropyl cellulose composite material as the controlled-release film. The micromorphology and the mechanical properties of the controlled-release film were investigated by scanning electron microscope (SEM) and tensile strength test. Moreover, the in vitro release behavior of the tablets in different media and the in vivo pharmacokinetics in Beagle dogs were also investigated. The results showed that the appearance of eroded controlled-release film had uniform micropores under SEM. Meanwhile, the eroded film had good mechanical properties to overcome deformation during the drug release process. The drug-release behavior of the prepared tablets fitted the osmotic-driven release mechanism. The pH values of media and food had no significant effect on the in vitro release. The pharmacokinetic results showed that the tablets had a steady drug concentration after oral administratio n with a lag time of 4 h, which indicated the products had a pulsatile controlled-release profile |
