| 英文摘要 | The success rate of porous conductive membrane preparation on fused silica capillary was greatly improved by control of voltage bias and electroosmosis effect. Basic buffer media was filled into the column, and a direct curvent voltage was applied between the capillary inlet and the etching reservoir during the etching process. The etching was stopped due to electroosmosis effect when electro-conductivity of the etching membrane occurred, and over etching was automatically avoided. The conductive membrane prepared was successfully used in on-line sample electro injection pre-concentration and capillary electrophoresis separation of model proteins of lysozyme and bovine serum albumin in 0.05 mol/L pH 2.50 Tris-H, PO, with 2% - 4% dextran, and a concentration factor close to the theoretical value was obtained. The difference of concentration factors for basic and acidic proteins was discussed. Detection limits for the model proteins were decreased to 1 x 10(-11) mol/L with a routinely used photometric detector (214 nm wavelegnth) by electro-injection pre-concentration for 30 min. |
