Pharmacokinetics of modafinil tablets in Chinese Mongolian, Uygur,and Hui healthy volunteers

| 英文摘要 | AIM To study the pharmacokinetics of modafinil tablets in healthy Chinese volunteers of Mongolian,Uygur,and Hui nationality.METHODS Thirty healthy Chinese Mongolian,Uygur and Hui volunteers（10 for every nationality,half males,half females） were given modafinil 200 mg,and the plasma samples were collected and determined by HPLC-UV method.The main pharmacokinetic parameters of modafinil and its metabolite modafinil acid in the three nationalities and different sexes were calculated with the DAS 2.0 practical pharmacokinetic program.RESULTS After a single oral dose of modafinil 200 mg,the main pharmacokinetic parameters of modafinil for Mongolian,Uygur and Hui nationality were as follows： t1/2（13 ± s 3）,（15 ± 4）,and（14 ± 2） h;tmax（2.2 ± 0.8）,（1.8 ± 0.5）,and（1.9 ± 0.9） h;ρmax（4.9 ± 0.7）,（4.8 ± 0.7）,and（4.7 ± 1.0） mg.L-1;AUC0-∞（69 ± 12）,（71 ± 16）,and（71 ± 20） mg.h.L-1,while the pharmacokinetic parameters of modafinil acid of the above three nationalities were： t1/2（5.2 ± 1.2）,（7.2 ± 0.7）,and（7.1 ± 1.8） h;tmax（2.9 ± 0.9）,（3.1 ± 1.3）,and（2.9 ± 1.1） h;ρmax（3.5 ± 0.8）,（2.8 ± 0.4）,and （2.8 ± 0.5） mg.L-1;AUC0-∞（34 ± 8）,（34 ± 6）,and（34 ± 4） mg.h.L-1.There were no significant differences observed（P 0.05） among the pharmacokinetic parameters between the three nationalities.Meanwhile,the pharmacokinetic parameters calculated according sex showed that the values ρmax（male（4.3 ± 0.5） vs.female（5.3 ± 0.7） mg.L-1,P 0.01） and Vd（male（65 ± 10） vs.female（51 ± 8） L,P 0.01）for modafinil and tmax（male（2.5 ± 0.9） vs.female（3.4 ± 1.1） h,P 0.05） and ρmax（male（2.8 ± 0.5） vs.female（3.3 ± 0.7） mg.L-1,P 0.05） for modafinil acid exhibited statistically significant differences.CONCLUSION Nationality has no significant impact on the pharmacokinetic parameters in healthy Chinese volunteers of Mongolian,Uygur,and Hui nationality after a single dose of modafinil tablets.However,there is some pharmacokinetic differences related to sex,which suggests possible requirements of dosage adjustments for sex difference. |
