Changes in the temperature tolerance profile of Gracilariopsis lemaneiformis from the perspective of photosynthesis, respiration, and biochemical markers after many years of vegetative propagation

英文摘要 | The temperature tolerance of Gracilariopsis lemaneiformis cultivated for nearly 35 years was compared to its wild-type counterpart after exposing them for 24-h to different temperatures (ranging from 9 to 29 degrees C), by surveying different parameters of photosynthesis and respiration, chlorophyll a fluorescence and oxidative stress damage/defense system, and the heat shock protein response. The performance index (PIABS) and the electron transport rate of the cultivated strains were significantly lower than those of the wild strains at 9 degrees C and significantly better at 29 degrees C. Additionally, the maximum quantum yield of PSII (F-V/F-M), as well as photosynthesis and respiration rates of the cultivated strain, was also significantly better than those of the wild strain at 29 degrees C. The malondialdehyde and H2O2 level of the cultivated strain were significantly lower than the wild strain at 29 degrees C. The ascorbate peroxidase (APX) and glutathione peroxidase (GSH-PX) activities, hsp70 expression level, and hsp70 protein content of the cultivated strain were higher than the wild strain at 29 degrees C. Damage resulting from membrane lipid peroxidation was evident in the wild strain at the high temperature, which decreased photosynthesis in the wild strain when compared to the cultivated strain. Based on the above outcomes, we conclude that cultivated G. lemaneiformis now has a weaker low-temperature tolerance and a greater high-temperature tolerance which may be related to long-term plant adaptation to the cultivation method. |
