天山雪莲指纹图谱建立与再生体系质量评价

中文摘要 | 本论文首次将指纹图谱的概念和方法引入到天山雪莲规模化组培快繁的工艺中,从表述多元化学组分和生物遗传学特性的化学指纹图谱和DNA指纹图谱两方面进行研究: 建立了天山雪莲的RP-HPLC指纹图谱,指认出13个共有峰中绿原酸、紫丁香苷、芦丁、木樨草素和芹菜素5种主要有效成分的特征指纹峰。实验证明该方法准确、重复性好,为天山雪莲的快速质量控制提供了依据。 采用HPLC法测定了天山雪莲组培过程中各阶段产物及人工种植实生苗中几种有效成分的含量,并与野生材料进行比较,对它们进行早期监测。结果表明移栽种植后的组培苗与实生苗色谱图与对照材料谱图大致相同,成分种类基本相似,其中绿原酸含量分别为0.84%,0.70%;芦丁含量分别为0.54%,1.80%,均高于《中华人民共和国药典》(2005版)的标准。 以天山雪莲种子苗、愈伤组织、丛生芽及再生植株的总DNA为模板,进行了RAPD反应条件的优化,分析了天山雪莲再生过程中DNA水平的变异。结果,20条随机引物共扩增出174条带,平均每条引物扩增8.7条带,其中多态性条带18条,多态性位点比率(P)为10.3%。天山雪莲再生体系与种子苗之间相似性系数为0.75-0.80,显示他们在DNA水平上发生了一定程度的变异。 本论文从化学水平和分子水平为组织培养和人工种植途径实现天山雪莲资源的可持续利用提供了依据。 |
英文摘要 | The concept and method of fingerprints were first introduced to the rapid regeneration system of Saussurea involucrata. The chemical fingerprints based on effective components and DNA fingerprints based on gene level were studied. Established RP-HPLC fingerprints of S . involucrata indicated 13 characteristic peaks, and 5 of them were confirmed to be chlorogenic acid, syringin, rutin, luteolin and apigenin. This method is simple and accurate with a good reproducibility and provides a reference standard for the quality control of S . involucrata. HPLC analysis method of effective component in S . involucrata regeneration system and seedling plant were established. The composition of effective components and fingerprints of S . involucrata regeneration plant and seedling plant after transplant were similar to those of wild plant. The contents of chlorogenic acid and rutin in S . involucrata regeneration plant and seedlings plant after transplant are markedly higher than those ruled by China pharmacopoeia (2005 edition). The seed plantlet, callus, cluster buds and regenerated plants of S . involucrata were used as the materials of extracting DNA. The RAPD reaction conditions were optimized, and the DNA level of variation by tissue culture were discussed. 174 bands were detected with 20 primers, with the average of 8.7 bands per primer. 18 polymorphic bands were detected among them, and the percentage of polymorphic loci (P) was 10.3%. The similarity coefficient between regeneration system and seed plantlet was in the range from 0.75 to 0.80, showing that there was some variations in DNA level. Results provided a reference for the tissue culture, artificial cultivation and sustaining usage of resource of S . involucrata. |
