英文摘要A W/O/W double emulsion-solvent diffusion process was developed for preparation of methoxypoly (ethylene glycol)-poly(DL-lactide)(PELA) microcapsule containing bovine hemoglobin (BHb), which is intended to be used as artificial red cells The influences of solidification method and type of organic solvent on oxygen-carrying activity of BHb were investigated. It was found that the preparative conditions such as stirring rate, type of organic solvent, type of stabilizer in outer aqueous solution, and solidification method had significant influences on the oxygen-carrying activity of BHb. When ethyl acetate was used as the organic solvent, PVA as the stabilizer in outer aqueous solution, and the double emulsion-solvent diffusion process adapted, the oxygen-carrying activity of BHb can be maintained if stirring rate was lower than 11000 r/min. On the other hand, the P50 and Hill coefficient of encapsulated BHb decreased when a high stirring rate (12000 r/min) was used. Increasing the volume of solidification solution could maintain the oxygen-carrying activity of BHb. The influence of preparative conditions on encapsulation efficiency and size of microcapsules were investigated. These included the polymer composition, the concentration of PELA, the concentrations of NaCl and stabilizer in the outer aqueous solution, the stirring rate for primary emulsification and re-emulsification, the volume of internal aqueous solution, the volume of re-emulsification solution. The relationship between encapsulation efficiency and size of microcapsules was also discussed theoretically. Microcapsule with a high encapsulation efficiency (90%) and a small diameter (3-5 u,m) was obtained when the PELA copolymer with a mPEG 2k block was used. The encapsulation efficiency and the size of microcapsule increased with the increase of PELA concentration. Increasing the concentrations of NaCl in outer aqueous solution resulted in the decrease of the diameter. As the concentration of stabilizer in outer aqueous solution increased from 5 g/L to 20 g/L, the size of microcapsule reduced while encapsulation efficiency increased, further increase of the stabilizer concentration lowed the encapsulation efficiency. Increasing stirring rate of the primary emulsification can improve the encapsulation efficiency though it had little influence on the size of microcapsule. The influence of stirring rate for re-emulsification was not apparent when a large volume of re-emulsification solution was used. However, when the volume of re-emulsification solution was small, the encapsulation efficiency and size of microcapsule decreased as the stirring rate for re-emulsification increased. It was also found that decreasing the volume of re-emulsification solution and increasing the quantity of wall polymer were advantageous for the preparation of microcapsules with small size and better encapsulation efficiency. Finally, a microcapsule with a relatively higher encapsulation efficiency of 50%, higher loading efficiency of 40%, smaller diameter of 1~2 u.m was obtained. The oxygen-carrying activity of BHb was well preserved. The prepared microcapsules containing bovine hemoglobin (BHb) using W/O/W double emulsion-solvent diffusion process and ethyl acetate as the organic solvent was intended to be used as the artificial red blood cells.
