褪黑素对光暗培养条件下紫锥菊毛状根生长和代谢的影响

中文摘要 | 褪黑素(Melatonin, MEL)是一种小分子吲哚类内分泌激素,在植物体内具有抗氧化、促进生长发育及昼夜节律等生理功能。本论文以紫锥菊毛状根为对象,研究了褪黑素对光照/黑暗条件下毛状根生长和代谢的影响,为深入理解褪黑素在植物的表达和调控提供了参考。 在连续光照和黑暗培养条件下,添加褪黑素能够明显提高紫锥菊毛状根中内源生长激素吲哚乙酸(IAA)的快速合成,从而促进了毛状根的快速生长。在连续光照培养条件下,紫锥菊毛状根中的超氧阴离子的产生速率、抗氧化酶 (超氧化物歧化酶、过氧化物酶和过氧化氢酶) 的活力高于黑暗培养的毛状根,添加褪黑素能够降低由连续光照产生的超氧阴离子的生成速率,并提高抗氧化酶的活力。添加褪黑素后,毛状根中花色苷、总黄酮、菊苣酸和总酚酸的生物合成减少,L-苯丙氨酸解氨酶活力下降。 从连续光照培养转向连续黑暗培养,紫锥菊毛状根内的MEL和IAA的含量明显升高,并维持在较高的水平。当毛状根从连续黑暗培养转向连续光照培养,毛状根内的MEL和IAA的变化规律与从连续光照转入连续黑暗培养的变化规律相反。在光暗周期培养条件下,毛状根内的MEL和IAA表现出波动性变化,从光周期转入黑周期过程中,MEL和IAA的水平逐渐增加,在暗周期结束时达到最高,从暗周期转入光周期后,MEL和IAA的水平逐渐降低,在光周期末期含量最低。当从光暗周期培养转入连续光照培养或者连续黑暗培养,MEL和IAA的周期性波动变化逐渐消失;当从连续光照培养或者连续黑暗培养再次转入光暗周期培养,MEL和IAA的周期性波动变化规律恢复。 |
英文摘要 | Melatonin has multiple functions as an antioxidant, growth promoter, photoperiodic regulator in plant. The objective of this study was to investigate the effect of melatonin on the growth and metabolism in the hairy roots of Echinacea purple L under light/dark conditions. Under both light and dark conditions, melatonin remarkably stimulated the hairy root growth due to the increase of IAA in the hairy roots by addition of melatonin. The activities of superoxide dismutase, peroxidase and catalase in the hairy roots treated by melatonin were higher than those in the hairy roots without melatonin treatment. The accumulation of anthocyanins, cichoric acid, flavonoids and phenolic compounds in the hairy roots treated by melatonin was lower than those in the hairy roots without melatonin treatment, and the activity of phenylalanine ammonium lyase was also decreased. When the hairy roots were transferred from constant light condition to constant dark condition, the accumulation of both melatonin and IAA increased to a high level. When the hairy roots were transferred from constant dark condition to constant light condition, the accumulation of both melatonin and IAA decreased to a low level. When the hairy roots were cultured under a photoperiod of 16 h-light/8 h-dark, melatonin and IAA levels in the hairy roots exhibited a remarkable photoperiodic response. Once the hairy roots grown under the light/dark regimes were transferred into the constant light or dark conditions, the photoperiodic response of both melatonin and IAA in the hairy roots disappeared gradually. When the hairy roots under the constant light/dark conditions were transferred into the photoperiod of 16 h-light/8 h-dark , the photoperiodic response of both melatonin and IAA in the hairy roots recovered again. |
