中文摘要溶液中蛋白质的絮凝沉淀是一复杂的多尺度的物理化学过程。研究结果使我们对蛋白质絮凝过程的认识更加深入和全面,并首次研究了蛋白质在絮凝过程中结构的变化。同时,研究结果表明,溶液化学环境对絮凝有显著的影响,不同的溶液环境对分子间的相互作用和絮粒聚集长大的影响是不一样的。絮凝剂的用量和分子量也是影响絮凝的重要因素。另外,通过显微镜直接观测了所得絮粒的物理形貌,从中可得到絮粒间黏结长大的信息。最后,我们提出了水溶性蛋白质的絮凝过程图解,描述了蛋白质絮凝的四个阶段。这一研究工作对于实现水溶性蛋白质的絮凝问题,合成和选择絮凝剂以及絮凝工艺条件的确定等具有一定的指导作用。
英文摘要In the present work, the mechanism of the flocculation of water-soluble protein using polymeric flocculant was studied and discussed. The different stages of flocculation in our system were investigated to understand the flocculation mechanism. The conformation changes of the protein in the flocculation process were studied using the Raman spectrophotometer. The results show that the second structure of protein was partly destroyed and the inner hydrophobic groups were exposed and combined with the hydrophobic parts of the polymeric flocculant through the hydrophobic action. At the same time, the complex coiled to form primary flocs. In this process, the entropy increased following the conformation changes of protein may be the inner driving force. On the other hand, effects of the solution chemical environments on the flocculation were investigated. The results show that the different solution chemical environments have the different effects on both molecular reaction and the formation of flocs. The effects of the dosage and molecular weight of flocculant as two important factors in the flocculation were also investigated. The effect of the dosage of flocculant was related to the salt concentration of the solution. Under the lower concentration of salt in the solution, the removal rate of protein increased linearly to the maximum with increasing the dosage of flocculant. But under the higher salt concentration, no molecular interaction occurred with increasing flocculant dosage. For a higher molecular weight (MW) PHP, the tendency of changes with increasing flocculant dosage was the same as lower MW PHP. But the flocculation of higher MW PHP was better than the lower MW PHP. The physical configuration of flocs was observed by the photomicroscope and SEM. It provided some information about the aggregation of flocs. A flocculation schematic diagram was presented to describe the flocculation process of water-soluble protein by polymeric flocculant. It contributes some to the understanding of flocculation of protein. And it is very important for the practical technique to solve the problem on the flocculation of water-soluble protein.
